Tryptophan is an essential nutrient in yeast that requires several preliminary reactions, each of which is catalyzed by a specific enzyme, before the final reaction yielding tryptophan can occur. If one gene is defective, the yeast will not be able to produce one of the necessary enzymes, thus stopping tryptophan production. Students will observe this biochemical pathway by controlling the nutritional environment and by crossing specific mutants. An incubator is recommended, but not required.
